Graphene: The future of cancer therapy

Cytotoxic agents are currently the main course of treatment for metastatic cancers. Unfortunately, these treatments often result in the destruction of healthy cells, are highly toxic and can lead to chemo-resistance. This support the need to find new effective targeted treatments and seek delivery methods which overcome drug resistance.

The exploration of graphene in drug delivery for cancer treatment was first initiated in 2008 by Stanford University and has been since the topic of intensive research. Due to the 2D structure and high specific surface area, graphene is able to adsorb a variety of therapeutic molecules with high capacity and even facilitate their entry into the cell. Moreover, by taking advantage of the enhanced permeability and retention (EPR) effect of tumors, the use of Graphene as a drug nanocarrier for cancer treatment has the potential to passively deliver chemotherapeutic and metabolism altering agents to cancer populations with increased specificity, whilst minimizing dosage and side effects of the drugs.

Manipulating the hydrophilic–lipophilic properties of graphene (blue hexagonal planes) through chemical modification would allow interactions with biological membranes (purple-white double layer), such as drug delivery into the interior of a cell (blue region) (Ref. doi:10.1038/nature11458 – A roadmap for graphene)

However, as most publications conclude, more pre-clinical studies and extensive clinical trials will be necessary to move this forward into the clinic to ensure patient benefit.

Towards this end, a research project led by Associate Professor Hanne Røland Hagland, is investigating the use of CealTech’s graphene as drug carrier for treatment of cancer, using cell models from leukaemia and colorectal cancer. The project is carried out at the Centre for Organelle Research (CORE), and is in collaboration with Prof. Lars Herfindal from the University of Bergen.

Ph.D. candidate Abdelnour Alhourani and MSc candidate Jan Lukas Førde are working on developing a graphene-based drug nanocarrier for dual chemotherapeutic and adjuvant metabolic drug delivery. Their work will focus on testing the cellular localization of fluorophore-conjugated graphene sheets of controlled sizes, and adapting appropriate biocompatible polymer functionalization to better understand the intracellular effects of graphene. The aim is to determine the capacity of CealTech’s graphene to enhance chemotherapy by shuttling adjuvant metabolism altering agents that are able to reverse chemo resistance. The viability of this approach is to be tested through multiples cell-based assays to monitor the cytotoxicity and metabolic shifts of cancer cells before additional testing in more advanced model systems.

Engineers in the U.K. unveil the world’s first graphene-skinned airplane

Graphene, the versatile miracle material that can be used for everything from creating better speakers for hearing aids to body armor that’s stronger than diamonds, has another application to add to its résumé. In the U.K., engineers at the University of Central Lancashire (UCLan) recently unveiled the world’s first graphene-skinned plane at the “Futures Day” event at Farnborough Air Show 2018. Called Juno, the 11.5-feet wide unmanned airplane also boasts graphene batteries and 3D-printed parts. The combination adds up to a pretty darn impressive whole.

“This project is a genuine world’s first,” Billy Beggs, UCLan’s Engineering Innovation manager, told Digital Trends. “It represents the latest stage of an ongoing collaborative program between academia and industry to build on innovative research, and exploit graphene applications in aerospace. We are establishing a lead in the industrial application of graphene.”

While the 3D-printed elements and graphene batteries are certainly exciting, the graphene-skinned wings are the most promising part of the project. Specifically, it is hoped that the use of graphene can help reduce the overall weight of the aircraft to increase its range and potential payload. This is made possible because the graphene carbon used in Juno is around 17 percent lighter than standard carbon fiber. Other properties of the graphene can help it counter the effects of potentially dangerous lightning strikes, due to its extreme conductivity, and protect the aircraft against ice buildup during flight.

Working with UCLan on the project is the Sheffield Advanced Manufacturing Research Center, University of Manchester’s National Graphene Institute, Haydale Graphene Industries, and assorted other businesses and research institutes.

“The U.K. Industrial Strategy highlights graphene as an example of a scientific discovery that needs to translate into industrial applications,” Peter Thomas, head of Innovation and Partnership at UCLan, told us. “Post-Brexit, the U.K. needs to continue to develop competitive advantage in aerospace through innovation.”

With Juno having made its stunning public demonstration, the next phase of the operation will include further tests to be carried out over the next two months. Should all go according to plan, airplanes such as this may well turn out to a particularly promising line of inquiry for graphene-related initiatives.

There’s a new way to 3D print graphene, the strongest material on Earth

With its incredible strength and potentially miraculous applications, there is a lot to be enthusiastic about when it comes to graphene. But it’s one thing to show off these possibilities in a lab; another entirely to turn it into something that’s usable in real-world situations. That’s something that researchers from Virginia Tech University and the Lawrence Livermore National Laboratory (LLNL) have been working to change. In the process, they have found a way of combining two of the most promising buzzwords in tech — “graphene” and “3D printing” — to open up a world of new exciting possibilities.

“We have been able to achieve 3D graphene aerogels and foams with arbitrary form factors and 3D features,” Xiaoyu “Rayne” Zheng, an assistant professor with the Department of Mechanical Engineering at Virginia Tech, told Digital Trends. “We formulated and printed light-sensitive graphene precursors that is compatible with a desktop SLA printer. This opens up freedom to realize 3D graphene with any topology co-optimized mechanical properties, hierarchical pore sizes, surface areas, [and] conductivities for a host array of applications.”

Regular graphene is a single layer of carbon atoms arranged in a honeycomb-style hexagonal lattice pattern. If graphene is packed, layer on layer, it becomes graphite: A material most commonly used as the “lead” in ordinary pencils. Now we love pencils as much as the next person, but anyone who has ever had a pencil lead snap on them may have a hard time believing this is one of the strongest materials on the planet. That’s because of the way that it is packed together, which fundamentally alters the structure of graphene.

The researchers on this project circumvented that by separating the individual sheets of graphene with air-filled pores, thereby allowing it to maintain its properties. The 3D-printable material that emerges at the end is something called graphene aerogel.

“Graphene aerogels are promising for a number of applications — including energy storage and conversion, catalysis, sorbents, and desalination,” Marcus Worsley, an LLNL researcher on the project, told us. “Recent work has shown some performance improvements for simple 3D-printed structures, but more complex, computer-generated architectures are predicted to be vastly superior. These gains should translate to devices that are more powerful, efficient, and longer lasting. This is the major thrust of our current and future work in this area.”

It may be some time before we are 3D printing with graphene in our home offices, but this still represents an enormous step in that direction.

“With regards to commercialization, we are always happy to work with potential commercial partners to bring our inventions to market,” Worsley continued. A paper describing the work was recently published in the journal Materials Horizon.

3D printing is one of the most exciting revolutions to hit manufacturing in decades, but it’s not without its limitations. Perhaps the biggest of these is the fact that, unless you create an unusually large 3D printer or break your model up into a whole lot of pieces, it’s difficult to create especially large 3D-printed objects. Fortunately, researchers from Singapore’s Nanyang Technological University have come up with a neat fix to the problem — and like all the best solutions, it involves a pair of robot arms.

In a new video demo, shown above, the researchers showcased how robots can build objects of virtually any size by moving around to carry out the printing. The demonstration employed off-the-shelf components, augmented by a nozzle, couplings, printing material and — perhaps most importantly — special guidance software developed at the university. It hints at a future in which whole teams of moving robots could collaborate to build structures.

“What we are seeing [in the video] is two mobile robots concurrently printing a concrete structure,” Pham Quang Cuong, Assistant professor of Mechanical and Aerospace Engineering at Nanyang Technological University, told Digital Trends. “The two robots first navigate from their base station to the printing site, then print by depositing a special type of cement layer by layer in a coordinated manner, and finally return to their base stations. The whole sequence was performed autonomously, [with] no human teleoperation.”

Cuong is no stranger to coming up with innovative uses for robots in construction tasks. Earlier this year, we wrote about another project he worked on in which robot arms were used to autonomously assemble a flat-packed Ikea chair — without having a massive argument over whether the instructions were being properly followed.

“The main potential use [for this latest project] is to print architectural, non-structural components, such as facades, either on-site or off-site,” he said. “Another team from our research center, the Singapore Center for 3D Printing, recently printed a bathroom unit using a similar technique.”

Regardless of where you live, houses are pretty expensive these days. That could change if the Italian 3D-printing company WASP (World’s Advanced Saving Project) has anything to say about it. The startup has just 3D printed a hut structure, called Gaia, using a combination of 3D-printed concrete and a mud-based material. And, of course, its giant, crane-based 3D printer. Total cost of the build? Around $1,000.

“The material consists of clay earth, rice straw, and rice husk,” Massimo Moretti, CEO of WASP, told Digital Trends. “The natural fibers allow [us to minimize] the shrinkage of the dry mixture and confer mechanical strength to the layered wall. By using the wet pan mill, the raw mixture has reached an interesting homogeneous plasticity that permits a good resolution in printed texture.”

In addition to the earth-and-rice-based material, concrete is used for the building foundations, while the roof is made of timber. The 215-square-foot build took around 10 days to complete, although more time would be required to furnish the place. It is also guaranteed that the cost would creep up north of the $1,000 price tag for a commercial build, since this only includes material costs and not labor.

“Gaia represents an example of reduced costs, especially if compared on the thermal performances that usually are obtained only with different traditional systems,” Moretti continued. According to WASP, Gaia does not require heating or air conditioning, and is able to maintain consistent temperatures indoors regardless of what time of year it is.

“At the moment, the company is evaluating every possibility to enter the construction market,” he said. “Probably the most reliable strategy will be a construction service, with our team involved in the wall construction.”

WASP isn’t the only group working in this area. Scientists from Estonia’s University of Tartu and the Estonian University of Life Sciences have developed a 3D-printable, concrete-style building material from milled peat, oil shale ash, and silica nanoparticles. Like Gaia, this material also promises to greatly reduce the cost of building new houses.

Still, if there’s anyplace where it’s good to have as many competitive products as possible in the mix, it’s building homes, quickly and cheaply, to house the world’s growing population.

When this army veteran broke the terms of his probation – and then lied about it – the judge knew that the guy had to serve his punishment. And so the judge threw him in jail.
